Basic Info

Chemically, the 1,3-diphenylacetone is C6H5CH2COCH2C6H5. The molecular structure of the 1,3-diphenylacetone is composed by a ketone group (-CO-), with phenyl groups (C6H5-) connected on both sides by methylene, creating asymmetric skeleton. The presence of aromatic rings in its structure gives it ketone-like properties and unique stability and reactivity. It is a common intermediate in organic synthesis.

Physically, the 1,3-Diphenylacetone is a white or pale yellow solid. It has a melting point of 34-36, and a boiling point around 331 under standard atmospheric pressure. It is soluble in organic solvents like ethanol, ether and benzene and can be dissolved uniformly. However, its solubility with water is very low and shows obvious hydrophobicity. The properties of the compound are stable at room temperatures due to the presence in the molecule of functional groups like benzene rings and ketone groups. Under conditions of high temperatures or strong oxidation, decompositions and oxidations may occur.

Dimethyl Disulfide

1,3-Diphenylacetone, a key organic intermediate, is used to build complex molecular structure. It has a wide range of applications and reaction mechanisms. Its reactive structure, formed by the methylene and keto groups (-CH2-), allows it to easily form an enolate ion under alkaline condition. This enables it to participate efficiently in condensation reactions. For example, under the catalysis of sodium ethoxide, it undergoes a cross-aldol condensation with benzaldehyde at 80-90degC for 2-3 hours, yielding 1,3,5-triphenylpentenone in over 75% yield. The product can be converted to polycyclic aromatics through cyclodehydrogenation, and is a precursor in the synthesis novel fluorescent materials. Condensation reactions with hydrazines produce pyrazoline-derived compounds, which are used to extract metal ions due to their excellent coordination property.

Due to its adaptable molecular structures, 1,3 diphenylacetone has been used as a synthetic base for many pharmaceutical intermediates. The Friedel Crafts reaction can modify its aromatic ring with functional groups like hydroxyls and aminos, and the keto group can then be converted into a hydroxyl group or methylene through reduction. This can control the compound’s biological activity and lipid solubility. Studies have shown that 3-phenyl-1-(4-hydroxyphenyl)acetone derivatives synthesized from it selectively inhibit cyclooxygenase (COX-2), with a half-maximal inhibitory concentration (IC50) of up to 12 mmol/L, making it a potential anti-inflammatory drug candidate. In the development of antibacterial drug, the derivatives can be combined with quinolone structure to enhance their inhibitory action against Gram-positive bacteria. This reduces the minimum inhibitory dose (MIC) down to 0.5mg/mL.

In materials science, the 1,3-Diphenylacetone has unique advantages for the preparation of polymer materials with functional properties. As a comonomer, it undergoes free radical copolymerization with methyl methacrylate (PMMA) initiated by azobisisobutyronitrile. The resulting copolymer, at a feed ratio of 1:4, has a glass-transition temperature (Tg), 15-20degC, higher than pure PMMA and a decomposition thermal temperature above 320degC. The conjugated aromatic ring is responsible for the increased molecular chain stiffness. This copolymer, when used to make coatings achieves a pencil-hardness rating of 3H with a grade one adhesion. The color difference (DE), after 1000 hours, is only 2.3, which is significantly better than conventional acrylic coatings. The ketone groups of this adhesive react with amines, forming cross-linked structures. This increases the shear strength to more than 8 MPa.

Chemical Research Field
The 1,3-diphenylacetone compound is a good model for studying the reactions of ketone compounds. In oxidation reactions, hydrogen peroxide can be used as an oxidant with sodium tungstate acting as a catalyst to selectively convert ketone groups into ester groups. The formation and transformation can be observed by tracking the changes in proton nuclear magnetic resonance (1H NMR spectrum) of the intermediate. This provides direct evidence to elucidate the nucleophilic mechanism of ketone addition. A comparison between sodium borohydride reduction mechanism and catalytic hydrogenation revealed that stereoselectivity of the former towards secondary alcohol was 3:1 while that of this latter was 1:2. This difference helps to understand the stereochemical properties in different reduction systems. The kinetics of phase transfer catalysis has also helped reveal the effect of charge transfer on reaction rates.
