Isoquinoline is an important nitrogen-containing heterocyclic organic compound with the chemical formula C9H7N. It is an isomer of quinoline and belongs to the quinoline family of compounds. It typically appears as a colorless to pale yellow oily liquid at room temperature and pressure, or condenses into a colorless crystalline solid at low temperatures (melting point approximately 26°C). It has a pungent odor similar to aniline, which becomes increasingly sharper with decreasing purity. Its density is approximately 1.099 g/cm³, slightly higher than that of water. It is insoluble in water but miscible in all proportions with a variety of organic solvents, including ethanol, ether, benzene, and chloroform. Its boiling point is approximately 243°C, and its refractive index ranges from 1.622 to 1.624. It is relatively stable at room temperature, but is somewhat sensitive to light and air. Long-term exposure can lead to slight oxidation and discoloration.
From a molecular perspective, the molecular skeleton of isoquinoline consists of a benzene ring fused to a pyridine ring at the ortho position, with the nitrogen atom located at the 2-position of the pyridine ring (adjacent to the benzene fusion site). This structure combines the aromatic stability of the benzene ring with the basicity of the pyridine ring (pKa approximately 5.4). There are two main routes for the industrial preparation of isoquinoline: one is extraction from coal tar. The quinoline fraction in coal tar can be isolated through fractionation, crystallization, and recrystallization, but its purity is relatively low (typically around 70%-80%), requiring further chemical purification to improve its quality. The other is synthetic production, such as using benzaldehyde and aminoacetaldehyde dimethyl acetal as raw materials, through cyclization and dehydrogenation reactions to produce isoquinoline. Alternatively, it can be prepared using the Bischler-Napieralski reaction, using β-phenylethylamine compounds as raw materials and cyclization in the presence of dehydrating agents such as phosphorus oxychloride. This synthetic method can produce isoquinoline products with a purity exceeding 95%, meeting the needs of high-end applications.

Isoquinoline’s excellent chemical activity, as well as its structural adaptability, brought about by its fused ring nitrogen structure, have established its core application value across multiple fields, including medicine, chemical engineering and scientific research. It is used as a “structural cornerstone”, in the construction of various clinical drugs. In chemical engineering, the compound serves both as a solvant and synthetic raw material. In scientific research, the compound provides support for testing, analysis and the development new materials. Its multidimensional potential makes it a multifunctional heterocyclic that links basic chemical research, pharmaceutical health and industrial production.
The basicity of its nitrogen atoms, and the possibility of modifying condensed ring structure have made isoquinoline a key core intermediate in pharmaceuticals. It is possible to introduce the nitrogen atom from the pyridine rings into the active groups through protonation and alkylation. The benzene rings can also undergo nitrations, halogenations, and other substitutions, which can optimize the structure of the drug molecules. It can be used to synthesize a variety of clinically useful drugs. For example, among alkaloid drugs it is the structural nucleus for opioid analgesics like morphine and acetaminophen, and it can control the analgesic effect and addiction by altering the condensed chain.
Its excellent solubility for organic compounds and polymer materials makes it a valuable solvent in chemical engineering. It can be used as a solvent for polymers such as epoxy resins and polyurethanes, enhancing their mechanical properties and improving the uniformity in the reaction system. As a synthetic material, isoquinoline can produce isoquinoline carboxylic acids through an oxidation reaction and prepare high-performance intermediates to synthesize azo dyes, which have bright colors, and anthraquinone, which are dyes that are fast and durable. It can be used to synthesize rubber vulcanization catalysts that will reduce the time required for rubber vulcanization and increase the heat and wear resistance of rubber products.
Isoquinoline is also indispensable in the field of scientific research. In analytical chemistry its alkalinity forms stable salts when combined with acidic substances. These are used as standard reagents in titration analyses or to purify and separate metal ions such as zinc, cadmium etc. This improves the accuracy of the analytical detection. In materials science, heterocyclic polymers containing nitrogen synthesized from the isoquinoline are important precursors to the development of organic semiconductors and high-temperature-resistant coatings because of their excellent conductivity.
